Sabouraud Dextrose Agar is a microbiological medium formulated with an enzymatic digest of casein, dextrose, and agar at an acidic pH, used for cultivation and enumeration of yeasts and filamentous fungi from clinical, environmental, and industrial samples.

The acidic reaction and high carbohydrate content favor fungal growth while restricting many bacteria; plates are typically incubated at 25–30°C for 24–72 hours, and the base medium may be supplemented with antibiotics to further limit bacterial contaminants when required.

  • 1.0% Tryptone
  • 4.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1.5% Agar
  • Cultivation of Yeasts and Molds: Supports robust growth of fungi from clinical, environmental, and industrial samples due to high dextrose and acidic pH.

  • Isolation of Candida spp.: Facilitates recovery and presumptive identification of yeasts from urine, stool, oral, and vaginal specimens.

  • Dermatophyte Recovery: Enables isolation of skin, hair, and nail pathogens such as dermatophytes from clinical specimens.

  • Environmental Monitoring: Used for air, surface, and personnel monitoring to detect fungal bioburden in cleanrooms and production areas.

  • Pharmaceutical Microbial Limits Testing: Applied in USP/EP/JP workflows for enumeration of yeasts and molds in nonsterile products.

  • Food and Beverage Testing: Detects and enumerates spoilage yeasts and molds in dairy, baked goods, juices, beer, and wine.

  • Fungal Contamination Screening: Screens cell culture reagents, process water, and raw materials for yeast and mold contamination.

  • Maintenance of Fungal Stock Cultures: Suitable for routine subculture and short-term maintenance of yeasts and filamentous fungi.

  • Selective Culture with Antibiotics: Used with additives (e.g., chloramphenicol, gentamicin) to suppress bacterial overgrowth during fungal isolation.

  • Teaching and Mycology Training: Standard medium for demonstrating fungal morphology and colony characteristics in educational labs.
