2XYT Top Agar is a semi-solid agar medium prepared from the 2×YT base for overlay plating applications.

It is used for bacteriophage plaque assays, lysogen screening, and formation of uniform bacterial lawns with E. coli hosts on compatible basal agar plates. The semi-solid consistency supports diffusion in overlays and consistent confluent growth during incubation.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 0.7% Agar
  • Bacteriophage Plaque Assays and Titration: Double-layer agar method to quantify and visualize plaques of coliphages (e.g., T4, T7, λ, M13) on Escherichia coli lawns.

  • Plaque Purification: Serial picking and re-plating of single plaques in soft agar overlays to isolate clonal phage populations.

  • Generalized Transduction: Soft agar overlays for P1-mediated transduction in Escherichia coli and P22 transduction in Salmonella enterica.

  • M13 Phage Rescue and Phage Display: Amplification of filamentous phage and library propagation using infected Escherichia coli in soft agar overlays.

  • Phage Host-Range and Spot Tests: Overlay lawns to assess susceptibility of hosts such as Escherichia coli and Salmonella enterica to different bacteriophages.

  • Blue/White Plaque Screening: X-gal/IPTG top-agar overlays with M13/λ systems to distinguish recombinant versus non-recombinant plaques on Escherichia coli.

  • Environmental Coliphage Enumeration: Detection and counting of coliphages from water or wastewater via double-layer agar overlays with Escherichia coli hosts.
