Tryptic Soy Agar is a general-purpose solid medium formulated with enzymatic digests of casein (tryptone) and soybean meal (soytone), sodium chloride, and agar. It is used for the cultivation, isolation, and maintenance of a broad range of nonfastidious bacteria, including laboratory strains of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

The peptone components supply peptides, amino acids, and nitrogen; sodium chloride helps maintain osmotic balance; and agar provides a stable surface for colony development and enumeration. Typical applications include routine culture propagation, colony morphology observation, environmental monitoring, and plate count methods in microbiological quality control.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 2.0% Agar
  • General Cultivation of Bacteria: Nonselective medium for robust growth of a wide range of organisms, including Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis.

  • Isolation and Colony Morphology: Streak plating to obtain isolated colonies and assess pigmentation, size, and texture.

  • Enumeration and Bioburden Testing: Colony counting from water, environmental, and process samples for microbial load assessment.

  • Environmental Monitoring: Settle plates, active air sampling, and surface contact plates for cleanroom and facility monitoring.

  • Stock Culture Maintenance: Routine subculture on plates or slants to maintain laboratory strains.

  • Purity Checks: Verification of culture purity and detection of contaminants in microbial workflows.

  • Food, Beverage, and Cosmetics Testing: Aerobic plate counts and microbial limits testing of raw materials and finished products.

  • Clinical and Research Sample Processing: Nonselective recovery of organisms from mixed or stressed samples prior to further testing.

  • Base for Enriched Media: Foundation for blood agar (e.g., addition of 5% defibrinated sheep blood) and other supplements.
