LB Low Salt Top Agar is a semi-solid LB-based overlay medium with reduced sodium chloride, used for bacteriophage plaque assays, transduction, and colony screening with E. coli hosts. The low ionic strength and soft agar consistency support standard overlay techniques on LB base plates.

Suitable for preparation at elevated temperature and use in overlays at approximately 45–50 °C. May be supplemented with divalent cations, antibiotics, or indicators as required by the experimental protocol.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 0.50% Sodium Chloride
  • 0.75% Agar
  • Bacteriophage Plaque Assays: Double agar layer plating to quantify phage titers on lawns of Escherichia coli (E. coli).

  • Phage Propagation and Amplification: Soft agar overlays to grow high-titer stocks of lytic phages on host bacteria.

  • Host Range and EOP Spot Tests: Overlay lawns to assess phage host range and efficiency of plating on strains such as E. coli and Salmonella.

  • Transduction Assays: P1 and P22 phage-mediated transduction experiments in E. coli and Salmonella using soft agar overlays.

  • Environmental Phage Detection: Enumeration of coliphages from water or environmental samples via double agar layer with E. coli hosts.

  • Phage Display Library Plating: Overlay plating for screening and titering recombinant phage display libraries.

  • Bacteriocin/Colicin Assays: Soft agar overlays to assess bacteriocin activity by inhibition zones on E. coli lawns.

  • Lysogeny Induction and Detection: Overlay-based assays to detect prophage induction and plaque formation.

  • Antibiotic/Compound Diffusion Spot Tests: Overlay lawns to visualize zones of inhibition against E. coli and related enterics.
