LB Agar in a Bottle is a general-purpose solid microbiological medium for routine cultivation and colony isolation of non-fastidious bacteria, including Escherichia coli. It provides nutrients and appropriate osmotic conditions for growth and colony formation on a firm agar surface.

Supplied in a bottle format for convenient preparation of plates or slants, it is suitable for maintenance and propagation of laboratory strains of E. coli and other enteric bacteria in standard cloning and microbiology workflows.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • Bacterial Cultivation and Isolation: Routine streaking and growth of Escherichia coli and other non-fastidious bacteria for single-colony isolation.

  • Plasmid Selection: Selection of transformants on antibiotic-supplemented plates to recover clones carrying resistance markers.

  • Blue-White Screening: Alpha-complementation with X-gal/IPTG to distinguish recombinant from non-recombinant colonies.

  • CFU Enumeration: Spread plating of serial dilutions to determine colony-forming units and viability.

  • Transformation Workflows: Recovery plating after chemical or electroporation-based transformations of Escherichia coli.

  • Replica Plating: Transferring colonies to selective conditions to screen auxotrophies or marker presence.

  • Phage Plaque Assays: Base medium for bacteriophage assays using soft agar overlays.

  • Conjugation and Transduction Selection: Selecting recipients after bacterial mating or P1 transduction on selective LB agar.

  • Colony Maintenance: Short-term storage of strains on plates, stabs, or slants (e.g., Bacillus subtilis).
