YPD Agar Plates with ClonNat-100 are a nutrient-rich yeast agar medium that supports growth of laboratory yeast strains, including Saccharomyces cerevisiae, and selection of nourseothricin-resistant clones. The presence of glucose provides a readily metabolizable carbon source for yeast growth, and 100 µg/mL clonNAT enables selection of cells carrying nat resistance markers (e.g., nat1/natMX).

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 2.0% Agar
  • 100µg/mL ClonNAT
  • Selection of NAT-Resistant Yeast Transformants: Isolates Saccharomyces cerevisiae colonies carrying NatMX or clonNAT-resistance cassettes on rich YPD (1.0% yeast extract, 2.0% peptone, 2.0% glucose, 2.0% agar) with 100 µg/mL ClonNAT.

  • Plasmid Maintenance and Recovery: Maintains yeast plasmids bearing the NAT selectable marker and enables recovery of individual clones.

  • Genome Editing and Integration Screening: Selects successful CRISPR/Cas9 edits or genomic integrations using NAT-marked donor cassettes.

  • Yeast Genetic Crosses: Selects haploids or diploids carrying NAT-marked alleles after mating, sporulation, or tetrad dissection.

  • Replica Plating and Strain Purification: Confirms genotype by transferring colonies onto NAT plates to verify marker retention and eliminate revertants.

  • Library and Fitness Screens: Enables pooled or arrayed yeast library selection (e.g., barcoded or deletion collections) under NAT pressure.

  • Colony Isolation and CFU Enumeration: Counts viable NAT-resistant yeast and isolates single colonies for downstream assays.

  • Competitive Growth Assays: Compares growth of NAT-tagged strains versus controls on rich medium with 100 µg/mL ClonNAT.
