YPD Agar Plates with ClonNat-25 is a nutrient-rich agar medium that supports the growth of yeast and the selection of nourseothricin-resistant strains. The inclusion of 2% glucose supplies a readily fermentable carbon source, and 25 µg/mL ClonNat (nourseothricin) enables selection of transformants carrying nat resistance markers.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 2.0% Agar
  • 25µg/mL ClonNat
  • Selection of NAT-resistant yeast transformants: Isolates Saccharomyces cerevisiae carrying natMX/NAT1 markers after transformation.

  • Plasmid/genome integration maintenance: Propagates strains harboring NAT-marked plasmids or genomic insertions.

  • CRISPR/Cas9 editing selection: Selects edited clones using natMX-based donor cassettes.

  • Yeast two-hybrid workflows: Maintains/selects Y2H strains when vectors or reporters use NAT resistance.

  • Colony isolation and purification: Streaks for single colonies and purifies NAT-resistant yeast lines.

  • Replica plating and genotype verification: Screens libraries or segregants on NAT to confirm marker presence.

  • CFU enumeration and viability checks: Quantifies viable NAT-resistant yeast under rich growth conditions.

  • Combinatorial selection: Used with additional antibiotics (e.g., G418, HygB) to select multi-marked strains.

  • Phenotypic screening under selection: Assesses growth phenotypes of NAT-marked mutants on rich medium.

  • Short-term strain maintenance: Routine passaging and storage of NAT-resistant yeast on 100 mm plates.
