YPD Agar Plates with Zeocin-250 are a nutrient-rich agar medium for the growth of yeast (e.g., Saccharomyces cerevisiae) and the selection of Zeocin-resistant strains. Glucose supports fermentative metabolism, and 250 µg/mL Zeocin (InvivoGen) provides selection for clones carrying the Sh ble resistance marker in yeast.

  • 2.0% Peptone
  • 1.0% Yeast Extract
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 2.0% Agar
  • 250µg/mL Zeocin® (an InvivoGen's product)
  • Selection of Zeocin-resistant yeast transformants: Isolating and maintaining Saccharomyces cerevisiae and Pichia pastoris (Komagataella phaffii) clones carrying Sh ble-based Zeocin® resistance.

  • Recombinant protein expression screening in yeast: Screening and stabilizing expression clones in S. cerevisiae or P. pastoris on rich medium with 250 µg/mL Zeocin®.

  • CRISPR/Cas9-edited yeast selection: Selecting edited colonies harboring Zeocin® resistance cassettes after genome engineering.

  • Vector maintenance and integration verification: Selecting integrative or episomal yeast shuttle vectors bearing Sh ble.

  • Colony isolation and replica plating: Picking single colonies and replica plating under Zeocin® pressure to verify stability.

  • Transformation efficiency and titering: Quantifying CFUs of Zeocin®-resistant yeast after transformation or electroporation.

  • Pichia pastoris expression strain workflows: Plate-based selection and maintenance for AOX1 and other systems using Zeocin® markers.

  • Library construction and screening in yeast: Maintaining and selecting pooled libraries (e.g., cDNA, mutant, or gRNA libraries) with Zeocin® resistance.

  • Contamination control during yeast enrichment: Suppressing sensitive microbial contaminants, including Escherichia coli and Staphylococcus aureus, while enriching resistant yeast.
