2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Carbenicillin-100 is a nutrient-rich agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The addition of 2% glucose enhances metabolic activity and biomass yield, and 100 µg/mL carbenicillin disodium salt enables effective selection of plasmid-bearing transformants.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • 2.0% Glucose
  • Selection of Carbenicillin-Resistant Clones: Growth and selection of Escherichia coli harboring β-lactam–resistant plasmids using 100 µg/mL carbenicillin.

  • High-Density Colony Growth: Rich 2xYT formulation supports robust colony formation for cloning and screening workflows.

  • Plasmid Maintenance and Amplification: Stable maintenance and amplification of ampicillin/carbenicillin-selectable plasmids in E. coli.

  • Reduced Leaky Expression: 2% glucose represses lac-based promoters, minimizing toxic protein expression during clone recovery.

  • Phage Display Workflows: Commonly used for M13 phagemid libraries (2xYT + glucose + carbenicillin) prior to helper phage infection.

  • Transformation Plating: Post-chemical or electroporation plating for efficient recovery and selection of transformants.

  • Colony Picking and Screening: Colony PCR, restriction analysis, and Sanger sequencing template preparation from selected colonies.

  • Starter Culture Initiation: Picking isolated colonies into liquid media for minipreps and downstream protein expression.

  • Library Screening: Plating and selection of combinatorial plasmid libraries in E. coli.

  • High-Throughput Automation: Large 245 mm low-profile format enables automated colony picking and trackable arraying.
