2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Ampicillin-100 is a nutrient-rich agar medium that supports the growth and selection of ampicillin-resistant E. coli strains.

The addition of 2% glucose enhances metabolic activity and biomass yield, and 100 µg/mL ampicillin sodium salt enables effective selection of plasmid-bearing transformants. Suitable for maintenance of recombinant strains, colony isolation, and routine cloning workflows.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 100µg/mL Ampicillin Sodium Salt
  • Selection and Maintenance of Ampicillin-Resistant Clones: Isolation and propagation of transformed Escherichia coli carrying Amp-resistant plasmids.

  • Molecular Cloning and Plasmid Propagation: High-yield colony growth on 2xYT (1.6% Tryptone, 1.0% Yeast Extract, 0.5% NaCl, 1.5% Agar) with 2.0% glucose and 100 µg/mL ampicillin for stable plasmid maintenance.

  • Colony Isolation and Picking: Efficient recovery of single colonies for downstream minipreps, PCR screening, and sequencing.

  • CFU Enumeration and Viability Testing: Quantification of ampicillin-resistant E. coli populations on selective solid media.

  • Library Construction and Screening: Plating of plasmid or genomic libraries under ampicillin selection for clone recovery and amplification.

  • Expression Workflow Support: Glucose-supplemented medium reduces basal expression from lac-based promoters during cloning steps in E. coli.

  • Phage/Phagemid Workflows: Plating of infected E. coli carrying ampicillin-selectable phagemids for titering and clone recovery.

  • Strain Banking and Stability Checks: Short-term storage and stability assessment of Amp-resistant E. coli strains on selective plates.
