2XYT (Yeast extract Tryptone) Agar with 2% Glucose, Carbenicillin-100, and Tetracycline-20 is a nutrient-rich agar medium that supports the growth and selection of carbenicillin- and tetracycline-resistant E. coli strains.

The addition of 2% glucose enhances metabolic activity and biomass yield, and 100 µg/mL carbenicillin disodium salt together with 20 µg/mL tetracycline hydrochloride enable effective selection of plasmid-bearing transformants carrying the corresponding resistance markers.

  • Dual-Antibiotic Selection of Escherichia coli Transformants: Selects for plasmids carrying beta-lactamase (Carbenicillin 100 µg/mL) and tetracycline resistance (20 µg/mL) on rich 2×YT (1.6% tryptone, 1% yeast extract, 0.5% NaCl) with 2% glucose and 2% agar.

  • Co-Transformation and Co-Maintenance of Dual-Plasmid Systems: Ensures retention of two compatible plasmids (CarbR + TetR) in E. coli for cloning, assembly, and pathway construction.

  • Reduced Basal Expression for Toxic or Burdensome Constructs: 2% glucose represses lac/T7-lac promoters, minimizing leaky expression in E. coli prior to induction.

  • Phagemid/Phage Display Workflows: Supports selection of CarbR phagemids and TetR helper/F′-associated elements in E. coli on rich 2×YT with glucose to control expression during phage production.

  • Verification of Recombinant Constructs Prior to Expression: Confirms plasmid integrity and antibiotic markers in E. coli before liquid culture and induction.
