YPD Broth is a nutrient-rich complex medium used for the cultivation and maintenance of yeasts, including Saccharomyces cerevisiae. It contains yeast extract, peptone, and anhydrous dextrose (glucose) as the primary carbon source.

This formulation provides amino acids, peptides, vitamins, and readily metabolizable carbohydrates suitable for routine propagation, starter cultures, and general laboratory applications involving yeast growth in liquid culture.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • Yeast Cultivation and Propagation: Rich, non-selective medium for robust growth of Saccharomyces and other yeasts (1.0% Yeast Extract, 2.0% Peptone, 2.0% Dextrose).

  • Starter and Overnight Cultures: Generation of high-density inocula for shake flasks, bioreactors, and fermentation studies.

  • Yeast Transformation Recovery: Post-transformation outgrowth to enhance viability and expression of introduced DNA.

  • Recombinant Protein Expression: Culturing engineered yeast strains for protein production prior to induction or harvest.

  • Plating and Colony Isolation (YPD Agar): Preparation of solid media for colony formation, streaking, and CFU enumeration.

  • Growth Curves and Fitness Assays: Optical density monitoring for doubling time, stress-response, and phenotype analyses.

  • Strain Maintenance and Glycerol Stocks: Routine passaging and preparation of freezer stocks for long-term storage.

  • Mating and Sporulation Pre-cultures: Pre-growth of haploid strains before transfer to selective or sporulation media.

  • Drug and Stress Tolerance Testing: Baseline rich medium for assessing effects of temperature, osmotic stress, or added compounds.

  • Library Amplification in Yeast: Expansion of pooled mutant or CRISPR-edited yeast libraries prior to selection.
