2XYT (Yeast extract Tryptone) Agar with Carbenicillin-100 is a nutrient-rich solid medium for the growth and selection of carbenicillin-resistant E. coli strains. Inclusion of 100 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants on plates.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt

  • Bacterial Transformation and Selection: Selects carbenicillin-resistant Escherichia coli transformants harboring beta-lactamase (bla/ampR) plasmids.

  • Plasmid Cloning and Maintenance: Supports robust growth of recombinant Escherichia coli for stable plasmid propagation on rich 2xYT with carbenicillin.

  • High-Copy Plasmid Amplification: Promotes rapid colony growth for downstream minipreps and maxipreps of ampR plasmids.

  • Colony Screening and PCR: Enables colony picking for colony PCR, restriction analysis, and Sanger sequencing workflows.

  • Protein Expression Clone Selection: Selects Escherichia coli expression strains carrying carbenicillin-resistant plasmids prior to induction.

  • Phage Display and M13 Work: Used to plate phagemid/helper phage-infected Escherichia coli requiring beta-lactam selection.

  • CRISPR and Library Screening: Selects edited or library-bearing Escherichia coli clones carrying ampR vectors.

  • Blue-White Screening (with X-gal/IPTG): Compatible with X-gal/IPTG supplementation for lacZ-based screening of recombinant colonies.
