2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Carbenicillin-100 is an agar medium for cultivation and selection of carbenicillin-resistant E. coli strains.

The medium includes 2% glucose as a readily metabolizable carbon source and 100 µg/mL carbenicillin disodium salt for selection of plasmid-bearing transformants.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 2.0% Glucose
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • Selective Plating of Escherichia coli with bla Plasmids: Carbenicillin (100 µg/mL) selects for beta-lactamase-bearing transformants on rich 2xYT agar.

  • Transformation Recovery and Colony Isolation: Supports robust outgrowth and discrete colony formation after heat-shock or electroporation.

  • Plasmid Maintenance and Propagation: Sustains high-copy plasmids under beta-lactam selection for reliable miniprep/maxiprep starter colonies.

  • Reduced Basal Expression in Lac-Based Systems: 2% glucose represses leaky expression prior to induction, improving stability of toxic or burdensome constructs in Escherichia coli.

  • Clearer Selection with Fewer Satellites: Carbenicillin’s stability minimizes satellite colonies compared to ampicillin, aiding accurate colony picking.

  • Colony PCR and Clone Screening: Enables rapid verification of recombinant colonies grown under selective pressure.
