2XYT (Yeast extract Tryptone) Agar with Kanamycin-50 is a nutrient-rich agar medium that supports the growth and selection of kanamycin-resistant E. coli strains. The inclusion of 50 µg/mL kanamycin sulfate enables effective selection of plasmid-bearing transformants.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • Selection of Kan-Resistant Transformants: Plate transformed Escherichia coli to select clones carrying kanamycin-marked plasmids using 2xYT nutrients (1.6% tryptone, 1.0% yeast extract, 0.5% NaCl) and 50µg/mL kanamycin in 1.5% agar.

  • Colony Picking and Screening: Isolate individual colonies for plasmid verification (mini-prep, restriction analysis, colony PCR, sequencing) from robust 2xYT growth with kanamycin selection.

  • Strain Maintenance and Streaking for Purity: Maintain and purify kanamycin-resistant Escherichia coli strains by streaking on selective 2xYT agar plates.

  • CFU Enumeration of Resistant Cells: Quantify viable counts of kan-marked cells following transformation, conjugation, or mutagenesis on selective 2xYT agar.

  • Recombinant Plasmid and Vector Propagation: Maintain plasmids carrying kanamycin resistance (e.g., expression or cloning vectors) in Escherichia coli prior to liquid culture scale-up.

  • Conjugation and Transconjugant Selection: Select kanamycin-resistant transconjugants after bacterial mating on nutrient-rich 2xYT agar.

  • Transposon Mutagenesis Selection: Recover and select Tn5/Tn-derived kanamycin insertions in Escherichia coli on 2xYT agar plates.

  • Library Plating and Screening: Plate and screen genomic, plasmid, or CRISPR libraries with kanamycin markers for high colony counts and even growth.

  • Phage/Phagemid Work (Overlay Base): Use as base plates for top-agar overlays when titering or rescuing kan-marked phagemids in Escherichia coli.
