2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Ampicillin-100 is a nutrient-rich agar medium that supports the growth and selection of ampicillin-resistant E. coli strains. The addition of 2% glucose provides a readily metabolizable carbon source, and 100 µg/mL ampicillin sodium salt enables selection of plasmid-bearing transformants.

Suitable for propagation of recombinant E. coli carrying ampicillin-selectable plasmids, colony isolation on solid medium, and maintenance of cultures under beta-lactam selection.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 2.0% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 100µg/mL Ampicillin Sodium Salt
  • Selection of AmpR Plasmid Transformants: Plating transformed Escherichia coli for selection of ampicillin-resistant colonies on rich 2×YT with 100 µg/mL ampicillin.

  • High-Density Colony Screening: Large 245 mm plates for library plating, replica plating, and colony lifts from AmpR E. coli clones.

  • Colony PCR and Sequencing Prep: Picking AmpR colonies from rich glucose-supplemented plates to verify inserts by PCR or Sanger sequencing.

  • Clone Maintenance and Streaking: Routine streaking and propagation of AmpR E. coli strains on a nutrient-rich, glucose-containing medium.

  • Plasmid Library Construction: Plating and screening of plasmid libraries requiring ampicillin selection on 2×YT agar.
