2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Ampicillin-100 is a nutrient-rich agar medium that supports the growth and selection of ampicillin-resistant E. coli strains. The addition of 2% glucose enhances metabolic activity and biomass yield, and 100 µg/mL ampicillin sodium salt enables effective selection of plasmid-bearing transformants.

Intended for cultivation, maintenance, and colony selection of recombinant E. coli on solid medium.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 100µg/mL Ampicillin Sodium Salt
  • Selection of Ampicillin-Resistant Transformants: Plating transformed E. coli to isolate colonies carrying Amp-selectable plasmids.

  • Plasmid Cloning and Maintenance: Maintaining recombinant plasmids in E. coli with 100 µg/mL ampicillin selection.

  • Colony Screening and Picking: Growing well-isolated colonies on rich 2xYT agar with 2% glucose for downstream analysis.

  • Colony PCR Templates: Generating robust colonies for rapid PCR-based insert verification.

  • Phage Display/Phagemid Workflows: Plating phagemid-containing E. coli; glucose helps repress lac-based promoters during selection.

  • Protein Expression Workflows (Pre-culture Selection): Selecting correct E. coli clones on plates prior to induction in liquid culture.

  • Strain Maintenance and Archiving: Streaking and short-term storage of recombinant E. coli under Amp selection.

  • Library Construction and Screening: Selecting diverse cloning libraries maintained on Amp-resistant E. coli backgrounds.

  • Transformation Efficiency Assessment: Quantifying colony-forming units after chemical or electroporation transformation.

  • Plasmid Stability Checks: Monitoring retention of Amp-selectable plasmids in E. coli colonies on rich selective agar.
