2XYT (Yeast extract Tryptone) Agar Plates with Carbenicillin-50 are nutrient-rich solid media that support the growth and selection of carbenicillin-resistant E. coli strains and the maintenance of beta-lactam–selectable plasmids.

Carbenicillin disodium salt at 50 µg/mL provides selection pressure against non-resistant cells, and the 2XYT base supplies peptides, amino acids, vitamins, and salts suitable for routine cloning, transformant screening, and colony isolation.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Carbenicillin Disodium Salt
  • Selective Plating of Transformants: Growth and selection of Escherichia coli carrying bla (ampicillin/carbenicillin)–resistance plasmids at 50 µg/mL carbenicillin.

  • Cloning and Subcloning Workflows: Isolation of single colonies on rich 2×YT agar for downstream plasmid construction and verification.

  • Colony Screening: Picking colonies for colony PCR, restriction analysis, or sequencing to confirm inserts.

  • Starter Culture Preparation: Selecting individual colonies to inoculate liquid media for minipreps, midipreps, and maxipreps.
