2XYT (Yeast extract Tryptone) Agar Plates with 2% Glucose, Carbenicillin-100, and Tetracycline-20 are nutrient-rich agar media that support the growth and dual-antibiotic selection of carbenicillin- and tetracycline-resistant E. coli strains. The addition of 2% glucose enhances metabolic activity and biomass yield, while 100 µg/mL carbenicillin and 20 µg/mL tetracycline hydrochloride enable selection of plasmid-bearing transformants.

Used for routine cloning and plasmid maintenance where beta-lactam and tetracycline resistance markers are required in E. coli.

  • Repression of Leaky Expression: 2% glucose supports catabolite repression to minimize basal expression from lac/T7 promoters during cloning and propagation steps.

  • High-Density Colony Growth: Rich 2×YT formulation (1.6% tryptone, 1.0% yeast extract, 0.5% NaCl) promotes robust colony formation for efficient clone picking and screening.

  • Phage Display and M13 Work: Plating and titering filamentous phage libraries on F′ Escherichia coli hosts with dual antibiotic selection.
