2XYT (Yeast extract Tryptone) Agar with 2% Glucose and Kanamycin-50 is a nutrient-rich agar medium that supports the growth and selection of kanamycin-resistant E. coli strains.

The addition of 2% glucose enhances metabolic activity and biomass yield, and 50 µg/mL kanamycin enables effective selection of plasmid-bearing transformants.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 50µg/mL Kanamycin
  • Kanamycin-Resistant Clone Selection: Isolation of transformants harboring kanamycin-selectable plasmids in Escherichia coli (50 µg/mL kanamycin).

  • Transformation Plating and Library Screening: Recovery and screening of colonies after chemical or electroporation-based DNA uptake.

  • Colony Isolation and Strain Maintenance: Streaking for single colonies and routine passage of kanamycin-marked strains on rich 2×YT medium.
