YPD Agar Plates with Zeocin-1000 are complex yeast media formulated for cultivation and selection of Zeocin-resistant yeast strains, including Saccharomyces cerevisiae. Glucose serves as the fermentable carbon source to support robust yeast growth on solid medium.

Zeocin® at 1000 µg/mL enables selection of transformants carrying a Zeocin resistance cassette (e.g., Sh ble), facilitating maintenance of plasmids or integration events in yeast under antibiotic pressure.

  • 2.0% Peptone
  • 1.0% Yeast Extract
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1000µg/mL Zeocin® (an InvivoGen's product)

  • Yeast Transformation Selection: Selects for Zeocin-resistant transformants carrying the Sh ble gene on YPD.

  • Clone Isolation and Purification: Streaking to single colonies to obtain pure Zeocin-resistant yeast clones.

  • Plasmid Maintenance in Yeast: Sustains selective pressure for episomal or integrative vectors in Saccharomyces and non-conventional yeasts.

  • Pichia (Komagataella) Integrant Screening: Identifies stable genomic integrants following transformation.

  • Titering and CFU Enumeration: Quantifies viable Zeocin-resistant yeast after transformation or library construction.

  • Recombinant Strain Construction: Builds and verifies engineered yeast strains prior to expression or fermentation.

  • Phenotypic Screening Under Selection: Assesses growth, stability, and segregation of traits with antibiotic pressure.

  • Cryostock Recovery and QC: Recovers and verifies identity of stored Zeocin-resistant yeast stocks.
