YPD Agar Plates with 1 M Sorbitol and Zeocin-500 are yeast extract–peptone–dextrose agar plates containing 1 M sorbitol for osmotic stabilization and 500 µg/mL Zeocin® (InvivoGen) for antibiotic selection. This medium supports cultivation of Saccharomyces cerevisiae and other yeasts while enabling selection of Zeocin-resistant transformants.

The sorbitol supplement provides osmotic support commonly used during yeast transformation and recovery, and Zeocin selection facilitates maintenance of constructs bearing Zeocin-resistance markers (e.g., Sh ble).

  • 2.0% Peptone
  • 1.0% Yeast Extract
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1M Sorbitol
  • 500µg/mL Zeocin® (an InvivoGen's product)
  • Yeast Transformation Recovery: Provides 1M sorbitol for osmotic stabilization of spheroplasts post-transformation or electroporation.

  • Selection of Zeocin-Resistant Yeast: 500µg/mL Zeocin® enables efficient selection of Sh ble–bearing transformants in Saccharomyces and Pichia workflows.

  • Clonal Isolation and CFU Enumeration: Supports colony formation on rich YPD (2.0% Peptone, 1.0% Yeast Extract, 2.0% Glucose, 1.5% Agar) while applying Zeocin® selection.

  • Plasmid Maintenance in Yeast: Sustains selective pressure for Zeocin®-marked episomal or integrative constructs during propagation.

  • Screening Recombinant Expression Strains: Enables picking and verification of Zeocin®-resistant clones for protein production (e.g., Pichia pastoris).

  • CRISPR/Cas9 Editing Workflows: Post-editing selection of edited yeast carrying Zeocin® resistance cassettes.

  • Spotting and Phenotypic Assays: Comparative growth, MIC, and fitness assays of Zeocin®-resistant versus control strains on rich medium.

  • Library Selections: Recovery and selection of mutant or gRNA libraries under Zeocin® pressure.

  • Streaking and Stock Maintenance: Routine streaking to maintain Zeocin®-resistant yeast lines and prevent loss of selectable markers.
