YPD Agar Plates with Hygromycin-250 are a complex yeast extract–peptone–dextrose medium solidified with agar and supplemented with 250 µg/mL Hygromycin-B. This formulation is intended for cultivation of yeasts, including Saccharomyces cerevisiae, on a rich carbon and nitrogen source.

The inclusion of Hygromycin-B at 250 µg/mL enables selection and maintenance of hygromycin-resistant transformants in yeast and other hygromycin-sensitive eukaryotic microbes.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 250µg/mL Hygromycin-B
  • Yeast Transformation Selection: Selects Saccharomyces cerevisiae transformants carrying hph/hygR markers after plasmid uptake or genomic integration.

  • Strain Maintenance and Propagation: Maintains hygromycin‑resistant yeast strains and plasmids on rich YPD for robust growth with selection pressure.

  • CRISPR/Cas9 Genome Editing Workflows: Screens edited yeast colonies using hygromycin‑B resistance cassettes for successful edits or integrations.

  • Yeast Two‑Hybrid and Display Systems: Selects for reporter or bait/prey constructs that include hygromycin resistance markers.

  • Genetic Crosses and Mating Assays: Recovers diploids or segregants retaining hygromycin markers after mating and sporulation workflows.
