2XYT (Yeast extract Tryptone) Broth is a nutrient-rich liquid medium that supports the cultivation of laboratory strains of E. coli and other facultative anaerobic enteric bacteria. Its composition provides readily accessible peptides, amino acids, vitamins, and minerals suitable for routine culture maintenance and scale-up.

This medium is commonly used for propagation of recombinant E. coli in cloning and expression workflows; selective agents (e.g., antibiotics) may be added as required for plasmid maintenance.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • High-Density Cultivation of E. coli: Rich medium supports rapid growth and elevated biomass for routine culture.

  • Plasmid DNA Amplification: Enhances yield in mini-/midi-/maxipreps from E. coli cultures.

  • Recombinant Protein Expression in E. coli: Supports robust expression in T7 and other bacterial systems.

  • Phage Display and M13 Propagation: Facilitates phagemid rescue and filamentous phage amplification from E. coli hosts.
