10X TBST, pH 7.4 is a Tris-buffered saline concentrate of 0.5 M Tris, 1.5 M sodium chloride, and 0.5% polysorbate 20 (Tween-20). Dilution to 1X gives 50 mM Tris, 150 mM sodium chloride, and 0.05% polysorbate 20 at pH 7.4. The 1 L fill makes 10 L of 1X buffer.

It is commonly used to wash plates in ELISAs and membranes in western blotting. Dilute 100 mL to 1 L with purified water.

  • 1.5M Sodium Chloride
  • 0.50% Tween-20
  • ELISA Plate Washing: Wash buffer concentrate that dilutes to 1X for removing unbound reagents between ELISA steps.
  • Western Blotting: Wash buffer concentrate for membrane washes in western blotting.
  • Immunohistochemistry and Immunofluorescence: Wash buffer concentrate for slide washes in IHC, ICC, and IF.
  • Antibody Dilution and Storage: Wash buffer concentrate, diluted to 1X, as a base for antibody dilution buffers.
  • Dot Blots and Protein Microarrays: Buffer for blocking, washing, and probing dot blots, slot blots, and protein microarrays.
  • ELISA Plate Blocking: Wash buffer concentrate, diluted to 1X, as a base for blocking buffers.
  • Hybridization and Nucleic Acid Blotting: Wash buffer concentrate for membrane washes in blot detection.
  • pH: 7.40 ± 0.04
