5M Tris-HCl, pH 8,0 is a concentrated buffering solution used to adjust and maintain pH in molecular biology and biochemistry workflows. At pH 8,0, it provides stable buffering capacity for procedures requiring mildly alkaline conditions.

Typical applications include preparation of working buffers such as TAE, TBE, and TE; pH adjustment of enzyme reaction mixes; nucleic acid extraction and purification steps; protein sample preparation; and column equilibration and wash buffers. The 5 M stock is commonly diluted to the desired working concentration before use.

  • 5M Tris-HCl
  • Polymerase Chain Reaction (PCR): Provides a concentrated stock to prepare PCR buffers at pH 8.0 for optimal polymerase activity.

  • Gel Electrophoresis: Used to formulate TAE/TBE and Tris-Glycine buffers for agarose gels and SDS-PAGE/native PAGE running systems.

  • Nucleic Acid Extraction and TE Buffers: Maintains pH during DNA/RNA extraction and in TE buffer for nucleic acid resuspension and storage.

  • Restriction Digests and Ligation: Serves as the Tris component in reaction buffers to stabilize pH for endonucleases and ligases.

  • Western Blotting: Prepares Tris-Glycine transfer buffer and TBS/TBST wash buffers for protein transfer and immunodetection.

  • Protein Purification: Forms the base of binding, wash, and elution buffers in ion exchange and affinity chromatography; supports dialysis buffers.

  • Enzyme Assays: Provides a stable pH 8.0 environment for kinetic measurements and activity assays.

  • Bacterial Cell Lysis and Transformation Buffers: Used in resuspension and lysis buffers for workflows involving E. coli and other bacteria.
