1 M Tris-HCl Solution, pH 7.4, is a concentrated buffering solution used to establish and maintain pH in biochemical and molecular biology workflows. The pH of 7.4 (measured at 25 °C) places it within the effective buffering range of Tris for applications requiring near-physiological conditions.

Common uses include preparing working-strength buffers such as Tris-buffered saline (TBS), adjusting the pH of assay and lysis buffers, and supporting nucleic acid and protein procedures including electrophoresis, chromatography, enzyme reactions, and sample preparation. The stock may be diluted to the desired working molarity with appropriate water or buffer.

Store tightly closed to minimize CO₂ absorption and pH drift; verify pH after dilution or temperature change as Tris pKa is temperature dependent.

  • 1M Tris-HCl
  • Polymerase Chain Reaction (PCR): Used in PCR reaction buffers to maintain optimal pH for polymerase activity.

  • Gel Electrophoresis: Component of Tris–Glycine and Tris–Tricine buffers for SDS-PAGE and native PAGE, ensuring stable pH during protein separation.

  • Western Blotting and Immunoassays: Preparation of TBS/TBST for membrane blocking and wash steps, preserving antigen–antibody integrity.

  • Protein Purification: Used in ion-exchange and affinity chromatography buffers to maintain stable pH during binding and elution.

  • Enzyme Assays: Provides consistent pH conditions for accurate enzyme kinetics and activity measurements.

  • Nucleic Acid Workflows: Included in buffers for DNA/RNA preparation and storage where a neutral pH is required.

  • Cell Lysis and Extraction: Common component of lysis buffers for tissues and microbial cells such as Escherichia coli.

  • Dialysis and Buffer Exchange: Maintains pH stability during sample dialysis and concentration steps.

  • pH: 7.40 ± 0.04
