Terrific Broth Agar Plates with Kanamycin-50 are nutrient-rich agar media that support the growth and selection of kanamycin-resistant E. coli strains. Glycerol serves as a carbon source, phosphate salts provide buffering capacity, and 50 µg/mL kanamycin enables effective selection of plasmid-bearing transformants.

  • 1.20% Tryptone
  • 2.40% Yeast Extract
  • 1.60% Glycerol
  • 1.25% Potassium Phosphate Dibasic
  • 0.23% Potassium Phosphate Monobasic
  • 1.50% Agar
  • 50µg/mL Kanamycin

  • Antibiotic Selection of Transformants: Isolates kanamycin-resistant Escherichia coli (E. coli) colonies carrying KanR plasmids (50 µg/mL kanamycin).

  • Plasmid Cloning and Maintenance: Supports growth and retention of KanR plasmids in Escherichia coli for routine cloning workflows.

  • Colony Screening: Enables colony PCR, sequencing prep, and restriction analysis from single kanamycin-resistant colonies.

  • Library Plating: Plating of transformation libraries (e.g., mutagenesis, combinatorial) under kanamycin selection to recover diverse clones.
