1 M Tris-HCl, pH 8.0 Solution is a laboratory buffer used to establish and maintain pH near 8.0 in biochemical and molecular biology workflows. Tris-HCl at pH 8.0 is commonly used for handling nucleic acids and proteins where a mildly basic environment is required.

Typical applications include dilution to prepare working-strength buffers for DNA and RNA workflows (e.g., TE components), electrophoresis systems (e.g., components of TAE), and protein purification or chromatography procedures requiring Tris-based buffering. The solution can be combined with salts or chelators as needed to formulate application-specific buffers.

  • 1M Tris-HCl
  • Polymerase Chain Reaction (PCR): Buffering component to maintain pH 8.0 for polymerase activity and DNA stability.

  • Agarose Gel Electrophoresis (DNA/RNA): Preparation of TE buffer and gel/loading buffers to stabilize nucleic acids during separation.

  • Protein Gel Electrophoresis (SDS-PAGE/native PAGE): Used in Tris–Glycine running and stacking/resolving gels to maintain consistent pH.

  • Western Blotting: Component of Tris–Glycine transfer buffers to support efficient protein transfer.

  • Protein Purification: pH control in ion-exchange and affinity chromatography buffers (e.g., equilibration, wash, elution).

  • Enzyme Assays: Provides stable pH 8.0 conditions for enzymatic kinetics and activity measurements.

  • Molecular Cloning: Base for restriction digestion, ligation, and transformation-compatible buffers.

  • Nucleic Acid Preparation: Formulation of TE (Tris-EDTA) for DNA resuspension, storage, and protection from degradation.

  • Plasmid DNA Workflows: Buffering in miniprep/maxiprep solutions for plasmid isolation from Escherichia coli cultures.

  • General Laboratory Buffering: Preparation of Tris-based buffers (e.g., TBS) and pH adjustment for biochemical assays.

  • pH: 8.00 ± 0.04
