10 mg/mL Tetracycline in 80% Ethanol is an antibiotic stock solution for selection and maintenance of tetracycline-resistance markers in bacterial culture. Tetracycline inhibits protein synthesis by binding to the 30S ribosomal subunit and is commonly used in cloning and expression workflows with E. coli.

Typical working concentrations are 10–20 µg/mL in bacterial media (adjust per protocol). Dilute into sterile aqueous media before use. Store protected from light at −20°C and avoid repeated freeze–thaw cycles.

  • Antibiotic Selection in Bacterial Culture: Selects for tetracycline-resistant transformants in E. coli and other bacteria during cloning workflows.

  • Plasmid Maintenance: Maintains plasmids bearing tet resistance markers in liquid culture and on plates for stable propagation in E. coli.

  • Preparation of Selective Media: Added to LB/agar and other media to create selective conditions for tet-marked constructs in E. coli, Bacillus subtilis, and Staphylococcus aureus.

  • Inducible Gene Expression (Tet Systems): Used as the regulatory ligand in Tet-On/Tet-Off systems for controllable gene expression in bacterial and mammalian cell culture.

  • BAC and Shuttle Vector Propagation: Supports selection and maintenance of tet-marked BACs and shuttle vectors during amplification in E. coli.

  • Screening and Counterselection: Enables differential growth to isolate desired clones and suppress tetracycline-sensitive background bacteria in mixed cultures.

  • Library Construction and Cloning: Provides selection pressure for combinatorial libraries and CRISPR/cloning constructs carrying tet resistance in E. coli.
