Tetracycline Hydrochloride is a broad-spectrum tetracycline-class antibiotic commonly used in microbiology and molecular biology workflows for the selection and maintenance of tetracycline-resistant bacterial strains, including E. coli. It acts by reversibly binding the 30S ribosomal subunit to inhibit protein synthesis.

This compound is water-soluble, light-sensitive, and typically used at working concentrations of 5–20 µg/mL in bacterial culture media. Stock solutions are commonly prepared in sterile water and stored refrigerated, protected from light, to maintain stability.

Applications include selection of plasmid-bearing transformants carrying tetracycline resistance markers and modulation of bacterial growth in routine laboratory procedures involving organisms such as E. coli.

  • Tetracycline Hydrochloride
  • USP Grade
  • Bacterial Selection and Plasmid Maintenance: Used as an antibiotic to select and maintain tet-resistance plasmids in Escherichia coli and other bacteria.

  • Inducible Gene Expression (Tet-On/Tet-Off): Regulates transcription in tetracycline-responsive systems in mammalian and microbial cells.

  • Antimicrobial Susceptibility Testing: Utilized in MIC determination and disk diffusion assays against organisms such as Staphylococcus aureus and Escherichia coli.

  • Mycoplasma Control in Cell Culture: Applied in treatment protocols to eliminate Mycoplasma contamination.

  • Protein Synthesis Inhibition Studies: Employed to inhibit bacterial ribosomes and mitochondrial translation for mechanistic studies.

  • Selective Media Supplementation: Added to agar or broth to create tetracycline-selective media for isolating or maintaining resistant strains.

  • Contamination Suppression in Mixed Cultures: Used to suppress unwanted bacterial growth in yeast or eukaryotic cell workflows involving Escherichia coli.

  • Stock Solution Preparation for Media: Prepared as sterile-filtered solutions for routine supplementation of culture media.
