Fluid Thioglycollate Medium Agar Plates are a thioglycollate-based solid medium for cultivation of aerobic, microaerophilic, and anaerobic bacteria. Yeast extract and tryptone provide peptides, amino acids, and vitamins; dextrose serves as a fermentable carbon source; sodium chloride maintains ionic balance; and agar supplies a solid growth surface.

Sodium thioglycollate and L-cystine help establish reduced conditions and an oxygen gradient conducive to anaerobic culture, while resazurin functions as an oxidation–reduction indicator that changes color in response to oxygen exposure, enabling visual assessment of the medium’s redox state.

  • 0.50% Yeast Extract
  • 1.50% Tryptone
  • 0.55% Dextrose
  • 0.05% L-Cystine
  • 0.25% Sodium Chloride
  • 0.05% Sodium Thioglycollate
  • 0.0001% Resazurin
  • 1.50% Agar
  • Sterility Testing (Pharma/Biologics): Used to detect aerobic and anaerobic contaminants in sterility and bioburden testing; reducing system (L-cystine/thioglycollate) and resazurin redox indicator support recovery of stressed microbes.

  • Isolation of Anaerobes: Primary cultivation of obligate anaerobes (e.g., Clostridium, Bacteroides, Fusobacterium) on 100 mm plates in anaerobic jars/chambers.

  • Oxygen Requirement Profiling: Differentiates obligate anaerobes, facultative anaerobes, and microaerophiles using the redox gradient and resazurin color change.

  • Clinical Microbiology Recovery: Recovery of fastidious or damaged anaerobes from wound, tissue, and abscess specimens (e.g., Clostridium perfringens, Bacteroides fragilis).

  • Environmental Monitoring (Cleanrooms): Settle/contact plating for anaerobic bioburden when incubated under reduced-oxygen conditions.

  • Food and Cosmetic Testing: Detection and enumeration of anaerobic contaminants in product and raw material testing.

  • Antimicrobial Screening (Anaerobes): Preliminary growth support for anaerobic isolates prior to standardized susceptibility testing workflows.
