This Tris-HCl buffer at alkaline pH is suited for nucleic acid handling, including DNA and RNA dissolution, dilution, and storage, as well as for preparing samples and reagents used in electrophoresis workflows. The controlled pH supports stability of nucleic acids during routine laboratory procedures and is compatible with common downstream enzymatic applications that operate in mildly basic conditions.

It can be used as a standalone buffer or as a component in combined formulations such as TE and electrophoresis buffers, and as a rinse or resuspension medium for plasmid DNA, PCR products, oligonucleotides, and competent cell preparations of E. coli. The solution is suitable for general molecular biology workflows requiring a low-ionic-strength buffering environment at basic pH.

  • 10mM Tris-HCl Solution
  • Polymerase Chain Reaction (PCR/qPCR): Provides a stable pH 8.5 environment for DNA polymerase performance and template dilution.

  • Reverse Transcription (RT): Maintains optimal pH during RNA dilution and cDNA synthesis setup.

  • Nucleic Acid Resuspension and Storage: Used to resuspend and store DNA or RNA, preserving integrity and stability.

  • Spin-Column and Gel Extraction Elution: Elutes DNA from silica membranes or gels with gentle, low-ionic-strength conditions.

  • Restriction Digests and Ligation Setup: Serves as a dilution buffer for enzymes and DNA to maintain consistent reaction conditions.

  • Library Preparation for NGS: Dilutes adapters, primers, and DNA fragments to ensure accurate molarity and pH control.

  • Enzyme Assays: Provides a defined pH for biochemical assays requiring Tris-based buffering.

  • Sample Normalization and Standards: Used to prepare calibration curves and control dilutions for quantitative analyses.

  • Buffer Component for Tris-Based Systems: Acts as a base buffer for preparing TAE/TBE variants or Tris-adjusted solutions.

  • Bacterial Lysate Preparation: Included in lysis and wash buffers for processing samples from Escherichia coli and other microbes.

  • pH: 8.50 ± 0.04
