Tryptic Soy Agar (TSA) Plates with Ampicillin-100 are a general-purpose agar medium for cultivation of nonfastidious bacteria and selection of ampicillin-resistant E. coli strains. The inclusion of 100 µg/mL ampicillin sodium salt provides selective pressure for plasmid-bearing transformants.

Suitable for colony isolation, routine maintenance, and CFU enumeration under ampicillin selection.

  • 1.5% Tryptone
  • 0.5% Soy Peptone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Ampicillin Sodium Salt
  • Selection and Maintenance of Ampicillin-Resistant Clones: Selects and maintains AmpR plasmid-bearing Escherichia coli after transformation.

  • Plasmid Cloning Workflows: Used for recovery plating and clone isolation following ligation, Gibson, or Golden Gate assembly.

  • Colony Isolation and Purification: Streaking to single colonies to obtain clonal AmpR strains for downstream work.

  • Colony PCR and Screening: Picking AmpR colonies for PCR, restriction analysis, or Sanger sequencing.

  • CFU Enumeration: Quantifying viable AmpR cells to assess transformation efficiency or library diversity.

  • Expression Screening: Evaluating expression constructs in AmpR Escherichia coli strains under selective pressure.

  • Maintenance of Reference Strains: Routine passaging of AmpR lab strains to prevent plasmid loss.

  • Contamination Control in Mixed Samples: Suppresses ampicillin-sensitive microbes, enriching target AmpR populations.

  • Phage and Host Workflows with Selection: Preserves AmpR plasmids in host lawns during bacteriophage assays using suitable AmpR hosts such as Escherichia coli.
