Tryptic Soy Broth is a general-purpose, nutrient-rich liquid medium for cultivation of a broad range of aerobic and facultative anaerobic bacteria. Enzymatic digests of casein and soybean meal supply peptides and amino acids; glucose provides a readily metabolizable carbohydrate; sodium chloride maintains osmotic balance; and dipotassium phosphate provides buffering capacity.

Common uses include propagation of laboratory strains such as Escherichia coli, Staphylococcus aureus, and Bacillus subtilis, preparation of inocula for downstream assays, and routine microbiological procedures in clinical, academic, and industrial settings.

  • 1.70% Tryptone
  • 0.30% Soytone
  • 0.25% Cerelose Anhydrous Dextrose (Glucose)
  • 0.50% Sodium Chloride
  • 0.25% Potassium Phosphate Dibasic
  • General Bacterial Cultivation: Rich, non-selective medium for robust growth of aerobes and facultative anaerobes such as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis.

  • USP Sterility Testing (Soybean-Casein Digest Medium): Employed per USP <71> for sterility tests of pharmaceuticals, biologics, and membrane filtration samples.

  • Bioburden and Microbial Limits Testing: Used as an enrichment and recovery broth for USP <61>/<62> microbial enumeration and specified microorganisms tests.

  • Environmental Monitoring and Swab Rinses: Recovery of low-level or stressed microbes from surfaces, equipment, and cleanroom EM swabs.

  • Inoculum Preparation: Routine propagation of cultures to generate standardized inocula for downstream assays (e.g., MIC setup, biochemical tests).

  • Growth Promotion Testing (GPT): Quality control of media performance using indicator strains including Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa.

  • Product and Device Recovery Studies: Extraction broth for microbial recovery in disinfectant efficacy, preservative efficacy (USP <51>), and medical device validation.

  • Food, Beverage, and Water Testing: Non-selective pre-enrichment and cultivation of diverse heterotrophic bacteria, including stressed organisms.

  • Blood Culture and Clinical Microbiology Workflows: Base medium for cultivating clinical isolates and for blood culture systems when appropriately supplemented.

  • Maintenance and Short-Term Holding: Temporary culture maintenance and subculturing prior to long-term storage or analysis.
