Tryptic Soy Broth (TSB) is a general-purpose, nutrient-rich liquid medium used for the cultivation and maintenance of a broad range of aerobic and facultatively anaerobic bacteria, including Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, and Salmonella enterica.

Enzymatic digests of casein and soybean meal supply peptides, amino acids, and growth factors; sodium chloride helps maintain osmotic balance; glucose provides a fermentable carbohydrate source; and dibasic potassium phosphate contributes buffering capacity. TSB is commonly used in sterility testing, microbial enumeration, and routine culture propagation.

  • 1.70% Tryptone
  • 0.30% Soytone
  • 0.50% Sodium Chloride
  • 0.25% Glucose
  • 0.25% Potassium Phosphate Dibasic
  • General Bacterial Cultivation: Broad-spectrum growth of nonfastidious organisms (e.g., Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis).

  • Sterility Testing: Detection of microbial contamination in pharmaceuticals, reagents, and medical devices.

  • Environmental Monitoring/Bioburden: Recovery of microorganisms from surface swabs, air sampling media, and rinse samples.

  • Enrichment Prior to Plating: Pre-enrichment of low-level contaminants (e.g., Salmonella enterica, Enterococcus faecalis) before selective agar plating.

  • Quality Control of Cultures: Routine propagation and maintenance of reference strains (e.g., ATCC) for QC assays.

  • Antimicrobial Susceptibility/Challenge Studies: Preparation of inocula and growth controls in efficacy testing of disinfectants and antibiotics.

  • Membrane Filtration Recovery: Growth medium for recovering stressed or filter-captured microbes from water and process samples.

  • Food and Beverage Microbiology: Enrichment of indicator and spoilage organisms from foods, ingredients, and in-process samples.

  • Clinical/Diagnostic Microbiology: Nonselective broth for primary culture and subculture of clinical isolates.
