10 mM Tris-HCl, pH 8.5 is a buffering solution for maintaining mildly alkaline conditions in molecular biology and biochemistry workflows. It is suitable for nucleic acid handling, including plasmid DNA preparation from E. coli, resuspension of DNA and RNA, and as a component in electrophoresis and blotting buffers where stable pH is required.

This solution is prepared with reagent-grade components and adjusted to pH 8.5 at room temperature. It is compatible with downstream enzymatic steps that require low ionic strength buffering and is commonly used for sample dilution, column equilibration, and routine laboratory procedures that specify Tris-HCl at pH 8.5.

  • 10mM Tris-HCl
  • Nucleic Acid Resuspension and Storage: Used to resuspend and store DNA or RNA at stable pH for long-term integrity.

  • DNA Elution (Spin/Gel Purification): Serves as an elution buffer for purified or gel-extracted DNA to maximize downstream enzymatic performance.

  • PCR/qPCR Template and Oligo Dilution: Maintains pH when diluting DNA templates, primers, and probes for amplification workflows.

  • Sanger/NGS Library Prep: Provides a low-salt, pH-stable medium for storing and diluting libraries, adapters, and barcodes.

  • Restriction/Ligation Workflows: Used to dilute DNA fragments and vectors without introducing inhibitory salts before enzyme reactions.

  • Protein Purification: Functions as a low-ionic-strength buffer for anion exchange equilibration, sample dilution, and gentle elution at pH 8.5.

  • Enzyme Assays: Supplies a defined pH environment for pH-sensitive enzymatic reactions and activity measurements.

  • Dialysis and Desalting: Employed as an exchange buffer to remove salts or small molecules while preserving biomolecule stability.

  • Plasmid DNA Handling: Used for resuspending and storing plasmid DNA isolated from Escherichia coli for reliable downstream use.

  • Electroporation-Compatible DNA Prep: Provides a low-conductivity buffer to prepare DNA for transformation while minimizing arcing risk.

  • pH: 8.50 ± 0.04
