1M Tris-HCl, pH 8.0, Endotoxin Tested, PCR Grade is a buffering solution for nucleic acid workflows. It is used to maintain pH 8.0 in applications such as PCR setup, DNA and RNA resuspension, and preparation of buffers for electrophoresis and cloning procedures.

The solution is tested for endotoxin and designated PCR Grade for use in molecular biology methods, including plasmid handling from E. coli and routine DNA/RNA processing steps requiring a defined pH environment.

  • 1M Tris-HCl
  • Polymerase Chain Reaction (PCR): Used in PCR reactions to maintain optimal pH conditions for enzyme activity.

  • Gel Electrophoresis: Provides Tris-based buffering for TAE/TBE and gel loading buffers to ensure consistent pH during DNA/RNA separation.

  • DNA/RNA Storage (TE Buffer): Forms the Tris component of TE buffer for resuspending and long-term storage of nucleic acids at pH 8.0.

  • Restriction Digests and Ligation: Maintains stable pH in cloning workflows, supporting restriction enzymes and DNA ligase activity.

  • Protein Purification: Used to make Tris buffers for ion-exchange and affinity chromatography, stabilizing protein structure during purification.

  • Enzyme Assays: Provides a consistent pH environment for accurate kinetic measurements and enzyme stability.

  • Western Blotting: Prepares Tris-Glycine(-SDS) running and transfer buffers, ensuring reliable protein separation and transfer.

  • TBS/TBST Preparation: Serves as the Tris base for Tris-buffered saline (with/without Tween 20) used in immunoassays and blot washing steps.

  • Cell Lysis Buffers: Constituent of Tris-based lysis buffers (e.g., RIPA) for protein extraction while maintaining pH 8.0.

  • Plasmid DNA Workflows: Used in neutralization and TE steps for plasmid preparation from Escherichia coli cultures.

  • pH: 8.00 ± 0.04
  • Endotoxin: ≤0.5EU/mL
