1 M Tris-HCl, pH 8.0, is a concentrated buffering solution for controlling and maintaining pH near 8.0 in molecular biology and biochemistry workflows. The solution is prepared at 1.0 mol/L and adjusted to pH 8.0 at 25 °C.

It is used to prepare and adjust Tris-based buffers, including TAE and TBE for nucleic acid electrophoresis, TE buffer for nucleic acid storage, and Tris-containing lysis, wash, and elution buffers for protein and enzyme applications. Use as supplied or dilute to the desired working concentration.

The pH of Tris buffers is temperature-dependent; verify pH at the intended working temperature. Store tightly closed and limit exposure to atmospheric CO₂ to minimize pH drift.

  • Nucleic Acid Electrophoresis: Component of TAE/TBE gel and running buffers for DNA/RNA gels, maintaining pH and ionic strength.

  • PCR and qPCR: Used to prepare and adjust reaction and dilution buffers, ensuring optimal pH for polymerase activity.

  • Restriction Digests and Ligation: Provides stable pH in enzyme buffers for restriction endonucleases and DNA ligases.
