1 M Tris-HCl, pH 6.5, is a pre-adjusted buffering solution used to maintain pH in biochemical and molecular biology workflows. Common applications include preparation of reaction buffers for enzyme assays, nucleic acid extraction and purification procedures, protein purification workflows, and electrophoresis systems based on Tris buffers.

At pH 6.5, it provides buffering in the mildly acidic range for processes requiring pH control below neutrality, and for dilution or adjustment of Tris-based buffer systems to defined conditions. It is routinely combined with components such as EDTA, salts, and reducing agents where chemically compatible, and is used in washing and handling steps for laboratory bacterial workflows, including work with E. coli.
