Tryptic Soy Agar (TSA) with Kanamycin-50 is a nutrient-rich agar medium that supports the cultivation of nonfastidious bacteria and the selection of kanamycin-resistant strains. The inclusion of 50 µg/mL kanamycin sulfate enables selection of plasmid-bearing transformants or resistant isolates in organisms such as E. coli.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • Selection of Kanamycin-Resistant Transformants: Isolating colonies harboring kanamycin-resistant plasmids (e.g., Escherichia coli, Bacillus subtilis, Pseudomonas putida).

  • Maintenance of Recombinant Strains: Routine passaging of engineered bacteria under 50 µg/mL kanamycin selection to preserve plasmids or genomic insertions.

  • Colony Screening for Cloning Workflows: Screening ligation/Gibson assemblies by growth on selective TSA to confirm acquisition of kan-marked constructs.
