Tryptic Soy Agar Plates with Kanamycin-25 are a nutrient-rich agar medium for cultivation of nonfastidious bacteria and selection of kanamycin-resistant strains.

The trypticase–soytone base supports the growth of organisms such as E. coli, and 25 µg/mL kanamycin enables selection and maintenance of kanamycin-resistant transformants and plasmid-bearing colonies.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 25µg/mL Kanamycin
  • Selective Isolation of Kanamycin-Resistant Bacteria: Tryptic Soy Agar (1.5% Tryptone, 0.5% Soytone, 0.5% Sodium Chloride, 1.5% Agar) with 25µg/mL kanamycin for isolating resistant strains such as Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa.

  • Plasmid Maintenance and Cloning: Maintains plasmids carrying kanR markers in hosts like E. coli during routine subculture and cloning workflows.

  • Transformation and Electroporation Screening: Post-transformation selection of kanamycin-resistant colonies from competent E. coli or other host strains.

  • Conjugation and Transconjugant Selection: Recovery and selection of kanR transconjugants in mating experiments (e.g., Agrobacterium tumefaciens to E. coli or other bacteria).

  • CFU Enumeration Under Selection: Quantification of colony-forming units from serial dilutions to measure frequencies of kanamycin-resistant cells.

  • Colony Purification and Streaking: Restreaking recombinant strains to ensure clonal purity while maintaining selection pressure.

  • Transposon Mutant Library Maintenance: Selection and propagation of Tn5/Tn-kanR mutant libraries in organisms such as E. coli or Salmonella enterica.

  • Verification of Genetic Constructs: Screening for correct insertion/maintenance of kanR-tagged cassettes, reporters, or expression vectors.

  • Strain Stability and Quality Control: Periodic plating to confirm retention of kanR plasmids or chromosomal markers in lab strains.

  • Environmental or Clinical Isolate Selection: Enrichment and isolation of kanamycin-resistant bacteria from mixed samples when selective pressure is required.
