Tryptic Soy Agar with defibrinated sheep blood is an enriched, general-purpose medium designed for the cultivation of a broad range of bacteria, including fastidious organisms. The nutrient base supplies peptides and amino acids, sodium chloride maintains osmotic balance, and the addition of sheep blood provides additional growth factors and enables visualization of hemolytic reactions.

This medium supports isolation and differentiation based on alpha, beta, or gamma hemolysis, and is commonly used for clinical specimens containing organisms such as Streptococcus pyogenes, Streptococcus agalactiae, Streptococcus pneumoniae, and Staphylococcus aureus. Intended for routine microbiological applications under appropriate laboratory conditions.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 5.0% Sheep's Blood
  • Cultivation and Isolation: General-purpose growth of clinical isolates, supporting both nonfastidious and many fastidious bacteria (e.g., Streptococcus spp., Staphylococcus aureus).

  • Hemolysis Detection: Differentiation of alpha, beta, and gamma hemolysis, aiding identification of Streptococcus pyogenes (beta) and Streptococcus pneumoniae (alpha).

  • Clinical Specimen Culture: Primary plating of throat, wound, and respiratory specimens to recover pathogens from mixed flora.

  • Antimicrobial Testing (Screening): Disk diffusion or qualitative susceptibility screening for organisms benefiting from blood supplementation.

  • Colony Morphology Assessment: Evaluation of colony size, hemolysis, and pigmentation (e.g., golden pigment of Staphylococcus aureus).

  • Satellite Test: Detection of Haemophilus influenzae via satellite growth around Staphylococcus aureus (X/V factor availability).

  • Quality Control and Verification: Routine QC of clinical workflows using reference strains (e.g., Streptococcus spp.) to confirm plate performance.

  • Enumeration from Mixed Cultures: Recovery and semi-quantitative assessment of bacterial load from environmental or clinical samples.

  • Aerobic/CO2/ Anaerobic Incubation: Versatile use under varied atmospheres to recover organisms requiring increased CO2 or reduced oxygen.
