Tryptic Soy Agar (TSA) Plates are a general-purpose solid medium for the cultivation, isolation, and enumeration of a broad range of non-fastidious microorganisms, including aerobes and facultative anaerobes from clinical, food, environmental, and industrial samples. The medium provides peptides, amino acids, and sodium ions to support microbial growth on a firm agar surface.

Typical applications include routine culture maintenance, purity checks, and environmental monitoring, with representative organisms such as Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • Routine Cultivation of Nonfastidious Bacteria: General-purpose growth of aerobes and facultative anaerobes such as Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Pseudomonas aeruginosa.

  • Isolation of Pure Cultures: Streak plating for colony isolation and morphology assessment.

  • Colony Enumeration (CFU): Spread plating and serial dilution plating for quantifying viable bacteria.

  • Environmental and Bioburden Monitoring: Recovery of aerobic heterotrophs from surfaces, air, water, and process samples.

  • Quality Control of Strains: Routine verification of colony morphology, pigmentation, and contamination checks.

  • Starter Cultures for Downstream Testing: Preparing colonies for biochemical assays, Gram staining, and molecular workflows.

  • Maintenance and Short-Term Storage: Holding working stocks of laboratory strains on plates.

  • Phenotypic Screening with Supplements: Selection or indicator-based screening when TSA is supplemented with antibiotics or dyes.

  • Replica Plating and Colony Picking: High-throughput transfer and picking for cloning and library maintenance.

  • Recovery of Stressed Cells: Cultivation of mildly stressed or sublethally injured bacteria after sample processing.
