Tryptic Soy Agar with Tetracycline-20 is a general-purpose agar medium used for cultivation and selection of tetracycline-resistant bacteria. The casein and soybean digests supply peptides and amino acids, and sodium chloride maintains osmotic balance.

Incorporation of 20 µg/mL tetracycline hydrochloride provides selection for tetracycline-resistance markers in organisms such as E. coli. Plates are supplied pre-poured and ready for inoculation.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 20µg/mL Tetracycline Hydrochloride

  • Selection of Tetracycline-Resistant Transformants: Plates transformants carrying tet-resistance plasmids (e.g., in E. coli) for cloning and recombinant strain recovery.

  • Plasmid Maintenance: Maintains selective pressure to retain tet-marked plasmids in laboratory strains during routine subculture.

  • Conjugation and Transposon Mutagenesis: Selects transconjugants or mutants bearing tetracycline resistance markers after mating or transposition.

  • Mixed Culture Selection: Suppresses susceptible background flora to isolate tetracycline-resistant colonies from environmental, food, or clinical samples.

  • Colony Screening After Transformation: Enables rapid picking and verification of recombinant colonies following chemical transformation or electroporation.

  • Strain Stability and Quality Control: Verifies retention of tet markers in production or research strains during passaging and storage.

  • Library Construction and Maintenance: Supports growth and preservation of genomic or plasmid libraries constructed with tet-selectable vectors.

  • Antimicrobial Resistance Surveillance: Enriches and enumerates tetracycline-resistant organisms (e.g., Escherichia coli, Staphylococcus aureus) from complex samples.
