Tryptic Soy Agar (TSA) Plates with Streptomycin-100 are a nutrient-rich agar medium that supports the cultivation and selection of streptomycin-resistant bacteria. The inclusion of 100 µg/mL streptomycin sulfate suppresses susceptible cells and enables growth of resistant populations, including laboratory strains of E. coli.

These plates are suitable for selection of streptomycin-resistant transformants or transconjugants and for routine maintenance of strains carrying streptomycin resistance markers.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Streptomycin

  • Selection of Streptomycin-Resistant Bacteria: Isolates colonies carrying streptomycin-resistance markers (e.g., aadA/strR) from mixed populations.

  • Screening Transformants/Transconjugants: Identifies successful cloning or conjugation events on plates selecting for streptomycin resistance.

  • Plasmid Maintenance: Maintains plasmids encoding streptomycin (or aadA streptomycin/spectinomycin) resistance in routine cloning workflows.

  • General Cultivation Under Selection: Supports growth and enumeration of non-fastidious organisms under antibiotic pressure, including Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, and Staphylococcus aureus.

  • Counterselection of Contaminants: Suppresses streptomycin-sensitive background flora to enrich for resistant strains in mixed cultures or environmental samples.

  • Antibiotic Resistance Surveillance: Recovers and quantifies streptomycin-resistant colonies from clinical, food, or water samples.

  • Phage and Bacteriocin Assays: Performs host-based assays while maintaining selection for streptomycin-resistant indicator strains.

  • Microbial Stock Preservation: Streaks and maintains laboratory strains requiring streptomycin selection for genetic stability.
