50 mg/mL Spectinomycin Solution is a sterile antibiotic stock for the selection and maintenance of spectinomycin-resistant bacterial strains. Spectinomycin, an aminocyclitol that targets the 30S ribosomal subunit, inhibits protein synthesis in susceptible Gram-negative bacteria and is commonly used to maintain plasmids carrying spectinomycin/streptomycin resistance markers (e.g., aadA) in hosts such as E. coli.

Typical working concentrations are 50–100 µg/mL in liquid or agar media. Store at −20 °C and minimize freeze–thaw cycles.

Bacterial Antibiotic Selection: Selects spectinomycin-resistant transformants in Escherichia coli and other Gram-negative bacteria carrying aadA/spectinomycin-resistance markers.
Plasmid Maintenance and Protein Expression: Maintains spectinomycin-selectable plasmids and expression vectors in Escherichia coli shake flasks and fermentations.
Dual-Selection Workflows: Enables co-transformation and stable maintenance of multiple plasmids using spectinomycin alongside other antibiotics (e.g., ampicillin, kanamycin).
Agrobacterium Vector Maintenance: Maintains binary and helper plasmids in Agrobacterium tumefaciens for plant transformation pipelines.
Cyanobacterial Genetics: Selects engineered strains of Synechocystis and Synechococcus using spectinomycin-resistance cassettes.
Chloroplast (Plastid) Transformation: Selects aadA-marked plastid transformants in plants (e.g., tobacco, Arabidopsis) during regeneration.
Algal Transformation: Selects aadA-marked nuclear or chloroplast transformants in Chlamydomonas reinhardtii and related algal systems.
Bacterial Conjugation Workflows: Applies counter/positive selection with spectinomycin to recover desired recipients or exconjugants during mating experiments.
