1 M Sodium Phosphate Buffer, pH 7.0, is a concentrated phosphate buffering solution used to establish neutral pH conditions for sample dilution, reagent preparation, and routine laboratory procedures requiring phosphate buffering capacity across molecular biology, microbiology, and biochemistry workflows.

Typical applications include pH stabilization in enzyme assays, nucleic acid and protein purification, chromatography equilibration and wash steps, and washing or resuspension of bacterial pellets such as E. coli. The 1 M stock is intended for dilution to working concentrations with sterile water or saline and may be sterilized by filtration or autoclaving according to protocol requirements.

1M Sodium Phosphate Buffer

Preparation of PBS and Phosphate-Based Buffers: Diluted to make working-strength phosphate-buffered saline (PBS) and related buffers for routine laboratory workflows.
Protein Purification: Used as equilibration, wash, and elution buffer in ion-exchange and affinity chromatography at controlled pH 7.0.
Enzyme Assays: Provides a stable pH 7.0 environment for enzymatic activity and kinetic measurements.
Protein Storage and Stabilization: Serves as a base for protein storage buffers to maintain protein integrity at neutral pH.
Dialysis and Buffer Exchange: Employed as a dialysis buffer to maintain constant ionic strength and pH during desalting or buffer exchange.
HPLC and FPLC Applications: Utilized as a non-volatile aqueous phase for protein and peptide separations requiring phosphate buffering.
Cell and Tissue Sample Preparation: Used in homogenization and wash steps to preserve physiological pH during sample processing.
Bacterial Culture Buffering: Added to growth media to stabilize pH during cultivation of Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa.
