2M Sodium Acetate, pH 4.5 is an aqueous buffering solution used to adjust and maintain mildly acidic pH in biochemical and molecular biology workflows. It provides acetate buffering capacity suitable for applications requiring controlled ionic strength and consistent pH.

Typical uses include nucleic acid precipitation workflows, sample desalting and cleanup steps, and chromatographic procedures that benefit from acetate buffering at pH 4.5. It can also be incorporated into protocols where a defined acetate environment supports reproducible assay conditions.

  • 2M Sodium Acetate
  • Nucleic Acid Precipitation: Added to ethanol or isopropanol precipitations to recover DNA and RNA efficiently using 2M Sodium Acetate, pH 4.5.

  • RNA Isolation and Enrichment: Acidic conditions favor RNA precipitation and help minimize DNA and protein carryover.

  • DNA Cleanup after Organic Extraction: Improves phase separation and DNA recovery following phenol–chloroform extraction.

  • Cation-Exchange Chromatography: Serves as an acetate buffer for protein and peptide separation at low pH.

  • Enzyme Reactions at Acidic pH: Provides stable buffering for acid-active enzymes (e.g., glycosidases, acid phosphatases).

  • Buffer Exchange/Dialysis: Used to transition samples into acetate buffer conditions prior to downstream workflows.

  • Precipitation of Contaminating Proteins: Low pH acetate conditions aid removal of proteins during nucleic acid prep.

  • Library Prep and Cleanup: Supports nucleic acid concentration steps in NGS and cloning workflows.

  • pH: 4.50 ± 0.04
