3M Sodium Acetate, pH 5.2 is an aqueous acetate buffering solution for laboratory use in molecular biology and biochemistry. It provides buffering capacity in the mildly acidic range suitable for workflows requiring acetate ions and controlled ionic strength.

Common applications include precipitation of DNA and RNA with ethanol or isopropanol, cleanup of nucleic acid preparations, preparation of acetate-based buffer systems for enzyme reactions and chromatography, and pH adjustment in sample processing.

Use by adding defined volumes to achieve the desired final salt concentration. Store tightly closed to minimize contamination and evaporation. For research and laboratory use.

  • Nucleic Acid Precipitation: Used with ethanol or isopropanol to efficiently precipitate and concentrate DNA and RNA from aqueous solutions.

  • Plasmid DNA Cleanup: Facilitates precipitation and cleanup of plasmid DNA following restriction digests, ligations, and enzymatic reactions.

  • Phenol–Chloroform Extraction: Provides acidic buffering during phenol–chloroform workflows to enrich and recover RNA.

  • Cation Exchange Chromatography: Serves as a low-pH acetate buffer for protein purification and elution on cation exchange resins.

  • Protein Crystallization: Functions as a buffering component near pH 5.2 in crystallization screens for optimizing protein crystal growth.

  • Small RNA Recovery: Enhances glycogen-assisted precipitation to improve yield of small RNAs and low-abundance nucleic acids.

  • DNA Concentration and Desalting: Enables precipitation-based concentration and removal of salts from DNA samples prior to downstream analyses.

  • HPLC Buffering: Acts as an aqueous-phase buffer for chromatographic separation of biomolecules requiring acetate buffering at pH 5.2.
